报告题目:Top-down Proteomics and PTMomics Using Isotopic Envelope Fingerprinting

摘要:Top-down proteomics and PTMomics (such as phosphoproteomcis and glycomics) are important branches of current cutting-edge systems biology. During the last 20 years, tremendous progresses in terms of both method development and application have been made in these two fields; tens of thousands of intact proteins and PTMs (such as phosphorylation and glycosylation) can now be routinely identified and quantified. However, lots of challenges still remain to obtain the complete molecular information of modified proteins (amino acid sequence, PTMs, PTM sites and structures). In this talk, I will give a brief introduction to our developments in top-down proteomics and PTMomics: (1) isotopic mass-to-charge ratio and envelope fingerprinting (iMEF) and ProteinGoggle for confident top-down protein characterization; (2) phospho-bracket (P-bracket) for unambiguous phosphosite localization of phosphopeptides; (3) database search engine GlySeeker for large-scale identification and visualization of N-glycans with full primary structure information.


田志新教授简介:2003年于中国科学院化学研究所获得博士学位。20042011年先后在美国明尼苏达大学和西北太平洋国家实验室做合作研究。20119月入选国家青年千人计划,被聘为中国科学院大连化学物理研究所研究员和高分辨质谱技术研究组组长。20133月被聘为同济大学化学系教授和博士生导师。目前主要研究领域是基于质谱的蛋白质组学和糖蛋白质组学。在完整蛋白质质谱分析、磷酸化位点准确定位、多糖拓扑结构鉴定、蛋白质解离通道、电喷雾生物大分子质谱原位解析及数据库搜索算法和引擎等研究领域取得了一定的研究成果。在Analytical Chemistry, Journal of Proteomics, Rapid Communications in Mass Spectrometry, International Journal of Mass Spectrometry, Chemical Reviews, Proceedings of the National Academy of Sciences of the United States of America, Angewandte Chemie-International Edition, Journal of the American Chemical Society, Genome Biology, Proteomics等刊物上发表论文50多篇。
